Omen13 is an experimental gene therapy candidate being developed by Gordian Biotechnology for the treatment of osteoarthritis (OA). It was identified using Gordian's proprietary in vivo mosaic screening platform, which utilizes Perturb-seq to evaluate genetic perturbations in disease models. Omen13 emerged as a top therapeutic candidate from a screen of 168 genetic perturbations in equine cartilage and synovium. In patient-derived chondrocyte-synovial co-culture assays, delivery of Omen13 via an adeno-associated virus (AAV) vector demonstrated the ability to increase glycosaminoglycan (GAG) production, promote chondrogenesis, and reduce the levels of pro-inflammatory cytokines, indicating potential for disease modification in OA.
