Overview

The Omicron KP.2-adapted monovalent COVID-19 vaccine is an updated mRNA vaccine formulation encoding the spike glycoprotein of the SARS-CoV-2 Omicron JN.1-lineage subvariant KP.2, designed to provide active immunization against currently circulating SARS-CoV-2 variants. It uses the same mRNA and lipid nanoparticle platform as earlier Pfizer–BioNTech (BNT162b2/Comirnaty) and Moderna (mRNA-1273/Spikevax) vaccines but replaces prior strains with a single KP.2 component, aiming to elicit neutralizing antibodies and cellular immune responses more specifically directed against KP.2 and related JN.1 sublineages.[1][2][5] Clinical data for KP.2-adapted BNT162b2 show robust neutralizing responses against KP.2, JN.1, and KP.3 with a safety and tolerability profile comparable to earlier monovalent and bivalent mRNA COVID-19 vaccines.[5] The formulation was developed as a seasonal 2024–2025 update by Pfizer/BioNTech and Moderna following FDA guidance to target JN.1 lineage, with KP.2 selected as the preferred strain to better match contemporary epidemiology.[1][2]
