Overview

**Omoprubart (CAN106)** is a novel, long-acting recombinant humanized monoclonal antibody of the IgG2-G4 κ subtype that specifically targets and neutralizes complement component 5 (C5), preventing its cleavage into C5a and C5b to inhibit formation of the membrane attack complex (MAC) responsible for cell lysis in complement-mediated diseases. Developed by CANbridge Pharmaceuticals, it acts downstream of C3 to preserve C3a and C3b for innate immunity while demonstrating dose-proportional pharmacokinetics, rapid and sustained C5 blockade, and favorable safety with only mild or moderate transient adverse events in clinical studies. Primarily in development for paroxysmal nocturnal hemoglobinuria (PNH), where Phase 1b data showed dose-dependent reductions in lactate dehydrogenase (LDH) up to 81%, hemoglobin increases up to 4 g/dL, and hemolysis control at extended four-week intravenous dosing intervals, outperforming frequent dosing needs of approved anti-C5 therapies in China.[1][2][3][6]
