Ompenaclid + FOLFIRI is an investigational combination regimen consisting of **ompenaclid**, a first-in-class oral inhibitor of the creatine transport channel SLC6A8, combined with the established chemotherapy regimen FOLFIRI, which includes folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan. Ompenaclid is being developed to target cellular energy metabolism in cancer cells, particularly in RAS-mutant colorectal cancer. FOLFIRI is a standard backbone for metastatic colorectal cancer that works by disrupting DNA synthesis and cell division through its component drugs. Ompenaclid is under evaluation for its ability to enhance the efficacy of FOLFIRI (and frequently bevacizumab) for patients with advanced/metastatic RAS-mutant colorectal cancer[1][2][5].
