OMS1620 is a small molecule antagonist of the melanocortin-2 receptor (MC2R), a GPCR for adrenocorticotropic hormone (ACTH). It is being developed for diseases associated with ACTH excess, in particular congenital adrenal hyperplasia (CAH). OMS1620 is designed to maximize receptor residency time, making it highly resistant to competition from high endogenous ACTH levels, supporting normalization of androgen levels in CAH patients while on physiological replacement doses of glucocorticoids. Preclinical data suggest best-in-class efficacy in both acute and chronic models of ACTH excess, with favorable effects on body and adrenal weight in animal studies. OMS1620 is currently in IND-enabling studies and has not yet entered clinical trials[1][2][3][5][8][11].
