Overview

OMS527 is an orally bioavailable, small molecule inhibitor of phosphodiesterase 7 (PDE7), developed by Omeros Corporation for the treatment of addiction and compulsive disorders, with a primary focus on cocaine use disorder and nicotine addiction. The drug acts by inhibiting PDE7, which modulates dopamine signaling in key brain regions involved in addictive and compulsive behaviors. This mechanism is distinct from current anti-addiction agents as it reduces craving and relapse without depressing the reward system or causing addiction itself. Preclinical studies have shown efficacy across multiple types of substance addictions (cocaine, opioids, nicotine, alcohol) as well as benefit in models of binge eating. Phase 1 clinical trials demonstrated that OMS527 was well tolerated with favorable pharmacokinetics supporting once-daily dosing[1][3][4][5][9].
