OMTX-707 is a preclinical antibody-drug conjugate (ADC) developed by Oncomatryx Biopharma that targets the endocytic receptor Endo180 (also known as MRC2) on tumor and stromal cells in solid tumors. In this construct, a human or humanized anti-Endo180 monoclonal antibody is site-specifically conjugated via a cleavable peptide linker (e.g., MC-GGFG–based) to a highly potent tubulin-inhibiting cytotoxic payload, yielding a drug–antibody ratio around 3.8 and low free antibody content.[9] Preclinical data in patient-derived xenograft models, including human leiomyosarcoma, show target-dependent binding to Endo180-expressing cells, efficient internalization, and robust antitumor activity with acceptable tolerability at doses up to at least 30 mg/kg in mice.[9] OMTX-707 is part of Oncomatryx’s tumor microenvironment–focused ADC pipeline and is being explored for the treatment of a range of solid tumors.
