OMV-WT (Wild-Type Outer Membrane Vesicles) are naturally occurring, nano-sized proteoliposomes secreted by Gram-negative bacteria, such as *Escherichia coli*. These vesicles are composed of a lipid bilayer containing various bacterial proteins, lipids, and pathogen-associated molecular patterns (PAMPs), most notably lipopolysaccharides (LPS). In the context of oncology research, OMV-WT is utilized for its intrinsic immunomodulatory properties, as it can activate the innate immune system primarily through the Toll-like receptor 4 (TLR4) pathway. This activation promotes the recruitment and infiltration of CD8+ T cells into the tumor microenvironment and stimulates the release of interferon-gamma (IFN-γ), which can inhibit tumor growth and potentially induce ferroptosis in cancer cells. OMV-WT serves as a foundational wild-type platform for genetic engineering to create targeted delivery systems, such as those designed to cross the blood-brain barrier for glioblastoma immunotherapy.
