ON104 is a fully human, bioengineered, Fc-silenced monoclonal antibody that specifically targets the oxidized isoform of Macrophage Migration Inhibitory Factor (oxMIF), a disease-related cytokine implicated in chronic inflammatory and autoimmune diseases. By neutralizing oxMIF, ON104 modulates disease progression and reduces inflammation without eliciting effector functions. Preclinical studies have demonstrated its efficacy in models of rheumatoid arthritis and glomerulonephritis, showing reduced clinical symptoms and tissue damage. The drug is designed for high-dose self-injection formulations to facilitate patient use at home. Developed by OncoOne, ON104 is being advanced for indications such as rheumatoid arthritis, nephritis (including glomerulonephritis), inflammatory bowel disease, systemic lupus erythematosus, and asthma[1][2][3][4][5][7][10].
