Overview

ON203 is a second-generation, bioengineered monoclonal antibody that targets oxidized macrophage migration inhibitory factor (oxMIF), a redox-dependent conformational isoform of MIF selectively present in tumor lesions and inflammatory sites. By binding to oxMIF, ON203 inhibits its activity, which is associated with poor prognosis in cancer patients. The antibody has been engineered for improved physicochemical properties, including reduced hydrophobicity and aggregation potential, increased plasma half-life, enhanced tumor accumulation, and superior antibody-dependent cellular cytotoxicity (ADCC) compared to first-generation anti-oxMIF antibodies such as imalumab. Preclinical studies have demonstrated that ON203 suppresses tumor growth in mouse models of prostate and colon cancer without acute toxicity. Developed by OncoOne, it is being advanced as an immunotherapy for solid tumors[1][3][4][5][6].
