Onartuzumab + paclitaxel is a combination therapy consisting of onartuzumab, a recombinant humanized monoclonal antibody targeting the MET receptor (also known as hepatocyte growth factor receptor), and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Onartuzumab inhibits the MET signaling pathway by binding to the extracellular domain of the MET receptor, thereby blocking its activation by hepatocyte growth factor (HGF). Paclitaxel disrupts cell division by stabilizing microtubules and preventing their depolymerization. This combination has been investigated in clinical trials for advanced solid tumors such as triple-negative breast cancer (TNBC) and non-small cell lung cancer (NSCLC). Clinical studies have not demonstrated significant improvement in progression-free or overall survival with this combination compared to standard regimens; increased rates of certain adverse events such as edema and hypoalbuminemia were observed in onartuzumab-containing arms[1][3][6][10].
