**Onartuzumab + pemetrexed + platinum** is an investigative combination regimen composed of: - **Onartuzumab**: a monovalent monoclonal antibody that binds to the extracellular domain of the MET receptor (also known as MetMAb or RO5490258). Onartuzumab inhibits hepatocyte growth factor (HGF) from binding to MET, thereby blocking the downstream MET signaling pathway involved in tumor cell proliferation, survival, and metastasis. - **Pemetrexed**: an antifolate small molecule antineoplastic agent that inhibits multiple enzymes involved in folate metabolism and DNA synthesis. - **Platinum**: refers to a platinum-based chemotherapy agent, either cisplatin or carboplatin, both of which induce DNA crosslinking and disrupt DNA synthesis and function leading to cell death. This combination has been studied in clinical trials for the first-line treatment of advanced (Stage IIIB/IV), non-squamous non-small cell lung cancer (NSCLC). According to available phase II data, onartuzumab demonstrated no significant improvement in clinical outcomes when added to standard pemetrexed and platinum chemotherapy[1][3].
