Onatasertib + toripalimab is a combination therapy consisting of onatasertib (ATG-008), a second-generation small molecule inhibitor of both mTORC1 and mTORC2, and toripalimab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. This combination is being evaluated for the treatment of various advanced solid tumors, including hepatocellular carcinoma and non-small cell lung cancer. The rationale for the combination is to leverage the synergistic effects of inhibiting the mTOR pathway, which is often dysregulated in cancer, while simultaneously blocking the PD-1/PD-L1 immune checkpoint to enhance the anti-tumor immune response. The TORCH-2 study, led by Sichuan University, specifically investigates this combination in patients with advanced solid tumors.
