**ONC-392 + docetaxel** is a combination currently being evaluated in phase 3 clinical trials for advanced non-small cell lung cancer (NSCLC) that is resistant to PD-1/PD-L1 inhibitor therapy. ONC-392 is a next-generation humanized anti–CTLA-4 IgG1 monoclonal antibody designed to selectively reduce regulatory T cells (Tregs) in the tumor microenvironment while preserving CTLA-4 protein recycling, thus enhancing antitumor activity and reducing toxicity compared to prior CTLA-4 antibodies. Docetaxel is a taxane-class cytotoxic chemotherapy agent that stabilizes microtubules, inhibiting cell division. The combination aims to synergistically enhance anti-tumor immune activity and cytotoxic effects in solid tumors, primarily NSCLC.
