ONC-784

ONC-784 is a bispecific antibody targeting the CD24-Siglec axis, designed for the treatment of solid tumors. Developed by AcroImmune (which merged with OncoC4 in 2024), the drug is part of a platform targeting a neo-CD24 epitope. CD24 is a potent 'don't eat me' signal overexpressed in approximately 70% of human cancers, which allows tumors to evade innate immune destruction by interacting with Siglec receptors (such as Siglec-10) on myeloid cells. By bispecifically targeting both CD24 and Siglec, ONC-784 aims to block this inhibitory signaling pathway and restore the phagocytic activity of macrophages and other innate immune cells against tumor cells. As of late 2024, ONC-784 was in IND-enabling studies, with clinical entry anticipated in 2025.
