ONC-841

ONC-841 is a humanized monoclonal antibody that acts as an antagonist of sialic acid-binding immunoglobulin-like lectin 10 (Siglec-10), an immune checkpoint molecule expressed on tumor-infiltrating immune cells. By blocking the interaction between Siglec-10 and its ligand CD24 on tumor cells, ONC-841 is designed to prevent tumor evasion of the immune system and rejuvenate anti-tumor activity in T cells, NK cells, and macrophages within the tumor microenvironment. This mechanism enhances phagocytosis of cancer cells and improves both innate and adaptive immune responses against tumors. Discovered by OncoC4, it is the first Siglec-10 antagonist to enter clinical development. The drug is currently being evaluated in a Phase 1 trial for advanced or metastatic solid tumors[1][2][5][6].
