ONC201 + nivolumab is a combination therapy that pairs two distinct anti-cancer agents. ONC201 (also known as TIC10 or dordaviprone) is a small molecule that inactivates AKT and ERK signaling pathways and activates the TRAIL pathway. It acts as a selective antagonist of dopamine receptor D2 (DRD2) and as an allosteric agonist of mitochondrial protease caseinolytic protease P (ClpP). Nivolumab (marketed as Opdivo) is an immune checkpoint inhibitor that targets PD-1 (programmed cell death protein 1). The combination was investigated to potentially overcome resistance to immune checkpoint inhibitors in microsatellite stable colorectal cancer, which typically does not respond to immunotherapy alone.
