Drug intelligence / Profile preview
Oncobax EH is a preclinical-stage live biotherapeutic product (LBP) consisting of a single bacterial strain of *Enterococcus hirae* (specifically strain 13144, or EH13144). Developed by EverImmune, a spin-off from the Gustave Roussy Cancer Center, Oncobax EH is designed as an oral adjuvant to cancer immunotherapy, particularly in combination with immune checkpoint inhibitors (ICIs) or chemotherapy (such as cyclophosphamide). The bacterium works by modulating the gut microbiota and enhancing the host's anticancer immune response. Mechanistically, *Enterococcus hirae* triggers systemic and intratumoral IFN-gamma-producing and CD137-expressing effector memory T-cell responses, and promotes dendritic cell activation via NOD2 and TLR2 pathways. Additionally, a bacteriophage-derived tape measure protein (TMP) antigen in *E. hirae* shares molecular mimicry with the tumor-associated antigen PSMB4, prompting cross-reactive CD8+ T-cell responses that target tumor cells. Oncobax EH is being investigated for the treatment of solid tumors, including breast cancer, lung cancer, and liver cancer.
