Overview

OncoD-F is an engineered oncolytic herpes simplex virus type 1 (oHSV-1) designed to express the asparaginyl hydroxylase factor inhibiting HIF (FIH1). Developed for the treatment of glioblastoma (GBM) and other solid tumors, OncoD-F specifically targets and downregulates Notch signaling within the tumor microenvironment. Notch signaling is frequently overactivated in GBM, contributing to tumor progression, therapy resistance, and an immunosuppressive microenvironment. By delivering FIH1 locally, OncoD-F inhibits Notch activity, which has been shown in preclinical models to reduce tumor growth, decrease tumor-associated macrophages, and increase T-cell infiltration. The virus combines direct oncolytic activity with gene therapy to modulate oncogenic signaling and enhance anti-tumor immunity.
