OncoFAP-11 is a small molecule radiopharmaceutical developed by Philochem AG, a subsidiary of Philogen. It consists of a high-affinity fibroblast activation protein (FAP) inhibitor conjugated to the radionuclide Lutetium-177 (177Lu) via a DOTAGA chelator. OncoFAP-11 targets FAP, a protease highly expressed in the stroma of various solid tumors but largely absent in healthy tissues. By binding to FAP with picomolar affinity, the compound delivers targeted beta-radiation to the tumor microenvironment, inducing DNA damage and cell death in both tumor cells and supporting stromal cells. It was identified using DNA-encoded chemical library (DEL) technology and has shown high tumor-to-blood ratios in preclinical models.
