Overview

**Oncolytic adenovirus armed with transforming growth factor-beta (TGF-β) blocker and interferon-gamma (IFN-γ)** is an engineered viral therapy designed to selectively replicate in and lyse tumor cells while counteracting the immunosuppressive tumor microenvironment (TME). The virus delivers a TGF-β blocker, such as a dominant-negative TGF-β receptor II (dnTGFβRII) or soluble TGF-βRII-Fc fusion protein, to inhibit TGF-β signaling that stabilizes regulatory T cells (Tregs) and suppresses effector T cell function, combined with IFN-γ expression to enhance Th1 immune responses, promote T cell fragility in Tregs, and boost antitumor cytotoxicity. This armed oncolytic virus induces tumor regression in resistant models like melanoma and pancreatic cancer by increasing IFN-γ production, T cell infiltration, and synergy with checkpoint inhibitors like anti-PD-1, with tumor-specific delivery minimizing systemic toxicity associated with TGF-β inhibition.[1][2][5]
