Drug intelligence / Profile preview
oncolytic herpes simplex virus type 1-mCherry (oHSV1-mCherry) is a genetically engineered, neuroattenuated oncolytic herpes simplex virus type 1 (oHSV-1) developed by researchers at the University of Padua. It features a genomic backbone similar to the clinically approved talimogene laherparepvec (T-VEC), with deletions in the γ34.5 and Us12 genes to reduce neurovirulence and enhance tumor selectivity. To further prevent replication in healthy neurons, the virus incorporates microRNA-124 (miR-124) target sequences downstream of the essential viral replication gene UL29. It also expresses the mCherry fluorescent reporter protein to allow tracking of viral infection and spread. oHSV1-mCherry is being investigated preclinically for the treatment of glioblastoma (GBM) and acute myeloid leukemia (AML), including studies evaluating its delivery to the central nervous system via carrier monocytes.
