Overview

PF-07263689 is a genetically engineered oncolytic vaccinia virus developed as an investigational therapy for solid tumors. It is designed to selectively infect and lyse tumor cells (oncolysis), leading to direct tumor cell death and the release of tumor antigens that stimulate both innate and adaptive immune responses against cancer. The virus has been further modified to express an IL-2 variant payload (IL-2gv), aiming to modulate the tumor microenvironment, potentially reverse resistance to immune checkpoint inhibitors, and induce killing of uninfected bystander tumor cells. The drug was administered intravenously in clinical trials for patients with locally advanced or metastatic solid tumors who had exhausted standard therapies. Development was discontinued after phase 1 trials due to business decisions rather than safety concerns[1][3][5].
