Oncorine

Oncorine is a genetically engineered oncolytic virus based on recombinant human adenovirus type 5. It was the first oncolytic virus product approved for cancer therapy worldwide and is marketed under the brand name Oncorine. Developed by Shanghai Sunway Biotech, it received approval from the Chinese State Food and Drug Administration (SFDA) in 2005 for use in combination with chemotherapy to treat head and neck cancers. The virus has deletions in the E1B-55kDa and E3 regions of its genome, which restricts its replication primarily to p53-deficient tumor cells—a common feature of many cancers—while sparing normal cells. Its mechanism involves direct lysis of tumor cells through selective viral replication as well as stimulation of anti-tumor immune responses by releasing tumor-associated antigens upon cell lysis. Clinical studies have shown that adding Oncorine to standard chemotherapy can significantly improve response rates compared to chemotherapy alone.
