Overview

ONCR-021 is an intravenously administered, selectively self-amplifying viral RNA therapy that encodes the genome of an optimized Coxsackievirus A21 (CVA21) strain and is formulated in a proprietary lipid nanoparticle to enable tumor-selective replication, oncolysis, and viral spread after delivery to tumor cells. It is being developed by Oncorus for solid tumors, with planned initial indications including non-small cell lung cancer, renal cell carcinoma, melanoma, and hepatocellular carcinoma. Preclinical studies support potent anti-tumor efficacy after IV dosing, including activity in multiple cancer models and tolerance in mice and non-human primates, and Oncorus had planned an IND submission in mid-2023 to begin clinical evaluation. The platform aims to achieve systemic delivery of an oncolytic virus via vRNA, allowing repeat dosing despite pre-existing neutralizing antibodies by producing oncolytic virions in situ within tumors.[1][3][4]
