Overview

ONCR-148 is an investigational oncolytic viral immunotherapy developed by Oncorus for the treatment of solid tumors. It is a recombinant herpes simplex virus type-1 (oHSV-1) engineered with neuron-specific microRNA (miR) attenuation of the ICP4 gene. This modification is designed to inhibit viral replication in healthy neurons, thereby reducing potential neurotoxicity, while preserving potent oncolytic activity within tumor cells. Furthermore, the ONCR-148 genome is engineered to express a transgene for a bispecific antibody targeting Fibroblast Activation Protein (FAP) and CD3. This bispecific antibody recruits and redirects the cytolytic activity of T cells toward FAP-expressing stromal cells within the tumor microenvironment, enhancing the overall antitumor immune response. Development of ONCR-148 was discontinued following the developer's financial restructuring and liquidation in 2023.
