ONCR-177 is an engineered recombinant oncolytic herpes simplex virus type 1 (HSV-1) designed for intratumoral administration in the treatment of solid tumors. It incorporates multiple safety features, including tissue-specific microRNA attenuation and a mutant UL37 gene to restrict replication and prevent neuropathic activity in normal cells. ONCR-177 is armed with five immunomodulatory transgenes: interleukin 12 (IL12), FLT3 ligand extracellular domain (FLT3LG ECD), CCL4, and antagonists to immune checkpoints PD-1 and CTLA-4. These modifications aim to stimulate both local and systemic anti-tumor immune responses while minimizing toxicity. Preclinical studies demonstrated potent oncolytic activity across various human cancer cell lines, selective replication in tumor cells, activation of dendritic cells, induction of tumor antigen-specific CD8+ T-cell responses, and abscopal effects in animal models. Clinical development has focused on advanced or refractory cutaneous, subcutaneous, or metastatic nodal solid tumors as well as liver metastases[1][2][4][6].
