**ONCR-788** is a synthetic viral RNA (vRNA) immunotherapy developed by Oncorus, consisting of an optimized Seneca Valley Virus (SVV) genome encapsulated in lipid nanoparticles (LNP) for intravenous administration. It selectively replicates in tumor cells, leading to oncolysis, immune stimulation via enhanced T-cell recruitment, PD-L1 upregulation, and macrophage polarization, with demonstrated preclinical efficacy in neuroendocrine tumors, small cell lung cancer, and other solid tumors, even in the presence of neutralizing antibodies. Planned for development as monotherapy or in combination with checkpoint inhibitors like anti-PD-1.
