ONCR-NEP is an investigational oncolytic virus therapy developed by Oncorus using its proprietary Nanoparticle Encapsulated Polynucleotide (NEP) platform. The therapy consists of a lipid nanoparticle (LNP) that encapsulates a DNA plasmid construct encoding the genome of the Seneca Valley Virus (SVV). Unlike traditional oncolytic therapies that deliver live viral particles, ONCR-NEP is designed for systemic (intravenous) delivery of the viral genome, which then initiates viral replication specifically within tumor cells. The SVV construct has been further engineered to express immunomodulatory payloads intended to recruit immune effector cells and enhance the adaptive immune response within the tumor microenvironment. It is primarily being developed for the treatment of neuroendocrine tumors and other solid malignancies.
