ONCT-808

ONCT-808 is an investigational autologous chimeric antigen receptor T (CAR T) cell therapy that targets the receptor tyrosine kinase-like orphan receptor 1 (ROR1) using a binding domain derived from zilovertamab. It is developed for the treatment of relapsed or refractory aggressive B-cell malignancies, including patients who have failed previous CD19 CAR T treatment. The product consists of a patient’s own T lymphocytes transduced with a lentiviral vector to express a CAR specific for ROR1. The mechanism of action involves immunologic cytotoxicity mediated by engineered T cells targeting and killing cancer cells expressing ROR1. Preclinical models demonstrated activity against multiple hematological malignancies and solid tumors; however, clinical development was discontinued following safety concerns and strategic business decisions[1][3][4][5].
