Onecet ER is a fixed-dose combination of tramadol hydrochloride and acetaminophen in a sustained-release (SR) formulation, developed by Daewon Pharmaceutical. Tramadol is a centrally acting synthetic opioid analgesic that exerts its effect through mu-opioid receptor agonism and the inhibition of norepinephrine and serotonin reuptake. Acetaminophen is a non-opioid analgesic and antipyretic that primarily inhibits prostaglandin synthesis in the central nervous system. This combination provides synergistic analgesic effects for the management of moderate to severe pain, such as chronic low back pain. The SR formulation is designed to provide prolonged pain relief with a reduced dosing frequency (typically twice daily) compared to immediate-release formulations, potentially improving patient compliance and maintaining stable plasma concentrations.
