**ONKT101** is an investigational allogeneic dual-targeted chimeric antigen receptor (CAR) natural killer (NK) cell therapy developed by ONK Therapeutics for relapsed/refractory B cell malignancies. It combines a CD19-directed CAR with a membrane-bound variant of tumor necrosis factor-related apoptosis-inducing ligand (TRAILv) that targets death receptor 5 (DR5) to enhance cytotoxicity and overcome resistance, leveraging NK cells' advantages over T cells such as lack of graft-versus-host disease and cytokine release syndrome risks. Manufactured using Avectas's SOLUPORE transient mRNA transfection technology for off-the-shelf repeat dosing, it is in first-in-human Phase 1 evaluation to assess safety and potential efficacy improvements over single-target CD19 therapies.[1][7][11]
