ONKT102 is a fully human, off-the-shelf, chimeric antigen receptor natural killer (CAR-NK) cell therapy candidate developed by ONK Therapeutics. It is designed with an optimized affinity CAR targeting CD38 and incorporates additional engineering features such as knockout of CD38 and CISH genes to enhance anti-tumor activity, persistence, and resistance to exhaustion in the tumor microenvironment. The therapy also includes a TRAIL variant (TRAILv) targeting DR5 to induce apoptosis in cancer cells. Its primary indication is for the treatment of relapsed or refractory multiple myeloma. Preclinical studies have demonstrated potent anti-tumor activity both in vitro and in vivo against CD38-positive multiple myeloma models[1][3][5][6][7][8].
