Overview

ONKT103 is a dual-targeted, off-the-shelf natural killer (NK) cell therapy developed by ONK Therapeutics. It is designed for the treatment of solid tumors and incorporates a chimeric antigen receptor (CAR) targeting tumor-associated MUC1 (TA-MUC1), as well as a TNF-related apoptosis-inducing ligand variant (TRAILv) that targets death receptor 5 (DR5). The CAR component specifically recognizes glycosylation patterns unique to cancer cells on TA-MUC1, while the TRAILv component engages DR5 to induce apoptosis via the extrinsic pathway. This dual mechanism aims to enhance tumor specificity and efficacy while reducing resistance and minimizing off-tumor toxicity. The therapy is currently in preclinical development for indications including ovarian cancer, breast cancer, and non-small cell lung cancer[1][2][3][4][6][7][8].
