ONKT105 is a highly functional, multi-gene edited allogeneic natural killer (NK) cell therapy candidate developed by ONK Therapeutics. It is designed for use either as a monotherapy or in combination with monoclonal antibodies or NK cell engagers. The therapy incorporates foundational gene edits such as Cytokine-inducible SH2-containing protein (CISH) knockout (KO), which enhances metabolic properties and improves in vivo persistence of NK cells, and may include additional edits like Transforming growth factor beta receptor 2 (TGFβR2) KO and knock-ins such as soluble Interleukin-15 (sIL-15) to further armor the NK cells against the tumor microenvironment. The primary indications under development are hematological malignancies and solid tumors. As of early 2024, ONKT105 remains in preclinical development[1][2][4].
