ONM-400 is an investigational, injectable, ultra pH-sensitive micellar delivery platform from OncoNano Medicine designed to encapsulate immunomodulatory biomacromolecules such as interleukin-2 (IL-2) and selectively release them within the acidic tumor microenvironment. By remaining quenched and stable at physiological pH and disassembling in response to the lower pH typical of solid tumors, ONM-400 concentrates IL-2 or other immune agonists in tumors while limiting systemic exposure, with the goal of enhancing antitumor immunity and reducing dose-limiting toxicity. Preclinical data presented at the Society for Immunotherapy of Cancer and AACR meetings showed high encapsulation efficiency, pH-dependent IL-2 release with preserved bioactivity, and prolonged, higher intratumoral exposure compared with free cytokine, supporting its development as a tumor-specific immunostimulatory platform.[1][5][6][9][10]
