ONM-405 is an investigational, tumor-acidosis responsive immunotherapy developed by OncoNano Medicine. It consists of Interleukin-12 (IL-12) encapsulated within the proprietary ON-BOARD pH-sensitive micelle platform. The drug is designed to remain stable and inactive at the neutral pH of systemic circulation (pH 7.4) but rapidly dissociates to release the IL-12 payload upon entering the acidic microenvironment (pH < 6.8) typical of solid tumors. This localized release mechanism is intended to activate cytotoxic T cells and natural killer (NK) cells directly within the tumor, thereby enhancing anti-tumor efficacy while avoiding the life-threatening systemic inflammatory response syndrome (SIRS) associated with systemic IL-12 administration.
