Overview

ONM-412 is a preclinical oncology drug developed by OncoNano Medicine. It consists of interleukin-12 (IL-12), a potent pro-inflammatory cytokine with well-established anti-cancer effects, encapsulated within an ultra-pH sensitive nanoparticle delivery system called ON-BOARD. This platform is designed to shield IL-12 from systemic exposure and selectively release it in the acidic tumor microenvironment, thereby improving tolerability and enhancing antitumor efficacy compared to unencapsulated IL-12 fusion proteins. Preclinical studies have demonstrated that ONM-412 induces tumor immune remodeling, increases infiltration of IFNγ+ and GzmB+ CD8 T cells and NK cells, and shows strong dose-responsive antitumor activity in animal models. The encapsulation technology also provides high stability, rapid recovery of IL-12 bioactivity upon acid activation, and reduced toxicity[1][2][3][4][6].
