ONM-421 is an experimental immuno-oncology agent developed by OncoNano Medicine, designed as a pH-activatable nanoparticle for the systemic delivery of a STING (Stimulator of Interferon Genes) agonist. Utilizing the proprietary ON-BOARD platform, ONM-421 consists of a STING agonist encapsulated within a pH-sensitive polymeric micelle. This delivery system is engineered to remain intact at the physiological pH of 7.4 but rapidly dissociate and release the agonist upon exposure to the acidic environment (pH < 6.8) characteristic of the tumor microenvironment. By targeting the STING pathway specifically within tumors, ONM-421 aims to stimulate the production of type I interferons and activate dendritic cells and T-cells, thereby inducing a potent anti-tumor immune response while avoiding the systemic cytokine storms often associated with non-targeted STING activation.
