ONO-3401

ONO-3401 is an orally bioavailable small molecule inhibitor of diacylglycerol kinase zeta (DGKζ), currently being developed by Ono Pharmaceutical. DGKζ is an enzyme that converts diacylglycerol (DAG) into phosphatidic acid, thereby acting as a negative regulator of T-cell receptor (TCR) signaling. By inhibiting DGKζ, ONO-3401 aims to maintain elevated levels of DAG, which enhances T-cell activation, proliferation, and effector functions. This immuno-oncology approach is intended to overcome the immunosuppressive tumor microenvironment and promote a more robust anti-tumor immune response. ONO-3401 is currently undergoing Phase I clinical evaluation for safety, tolerability, and pharmacokinetics in healthy volunteers and patients with solid tumors.
