Overview

ONO-4053 is a small molecule drug developed as a selective antagonist of the prostaglandin D2 receptor 1 (DP1). It was designed to block the action of prostaglandin D2 (PGD2), which is primarily produced by mast cells and contributes to nasal symptoms such as congestion, sneezing, rhinorrhea, and itching in allergic rhinitis. Clinical trials demonstrated that ONO-4053 had greater efficacy than pranlukast (a leukotriene receptor antagonist) in improving nasal symptoms associated with seasonal allergic rhinitis, with a similar safety profile. The drug was developed by Ono Pharmaceutical for oral administration but its global development status has been discontinued[1][4][9].
