ONO-4474

ONO-4474 is a novel, orally available, peripheral-specific pan-tropomyosin receptor kinase (pan-Trk) inhibitor developed for the treatment of pain, particularly musculoskeletal pain associated with osteoarthritis. It selectively inhibits tropomyosin receptor kinases TrkA, TrkB, and TrkC with low penetration across the blood-brain barrier. The drug acts as an antagonist to these receptors, thereby modulating nerve growth factor (NGF)-mediated nociceptive signaling and providing analgesic effects. Clinical studies have shown that ONO-4474 can reduce pain in patients with moderate-to-severe osteoarthritis of the knee who have an inadequate response to NSAIDs.
