ONO-4817 is a **small molecule inhibitor of matrix metalloproteinases (MMPs)**, specifically inhibiting MMP-2, MMP-3, MMP-9, MMP-12, and MMP-13, but not MMP-1 or MMP-7[1][3][8][9]. The drug exhibits potent inhibition of MMP-2 and MMP-9 (IC50 ~0.5-1.1 nM), which are enzymes that play important roles in the degradation of extracellular matrix and are implicated in tissue remodeling, tumor invasion, angiogenesis, and inflammation[1][2][5][7][8]. ONO-4817 binds reversibly to the zinc-binding region of target MMPs[5]. Developed by Ono Pharmaceutical, ONO-4817 showed therapeutic benefit in animal models of cancer metastasis, peritoneal fibrosis, and arthritis, but was discontinued in clinical development[2][3][5][10].
