ONO-7018

ONO-7018 is an orally bioavailable, potent, and selective small molecule inhibitor of mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1). MALT1 is a key component of the CARD11-BCL10-MALT1 signalosome complex that activates nuclear factor-kappaB (NF-κB) signaling in response to B-cell or T-cell receptor stimulation. This pathway promotes survival and proliferation in B-cell and T-cell lymphomas. By inhibiting MALT1, ONO-7018 disrupts this signaling cascade, potentially exerting antitumor effects against malignancies such as non-Hodgkin lymphoma (NHL) and chronic lymphocytic leukemia (CLL). The drug has demonstrated preclinical efficacy in several lymphoma models and is currently being evaluated in Phase 1 clinical trials for relapsed or refractory NHL and CLL[1][2][3][4][5].
