Overview

ONO-7475 is an orally bioavailable small molecule inhibitor of the **TAM** (Tyro3, AXL, and MER) family of receptor tyrosine kinases, with high potency against **AXL** and **MER**. Developed by **Ono Pharmaceutical**, the compound is primarily being investigated for its ability to overcome or delay resistance to EGFR tyrosine kinase inhibitors (TKIs) in patients with non-small cell lung cancer (NSCLC). AXL signaling is a well-characterized bypass mechanism that mediates acquired resistance to drugs like osimertinib. By inhibiting AXL and MER, ONO-7475 aims to restore sensitivity to EGFR-targeted therapies and improve clinical outcomes in EGFR-mutated, advanced, or recurrent NSCLC. It is currently undergoing Phase I clinical evaluation in Japan.
