ONP-302 is a biodegradable nanoparticle composed of poly(lactic-co-glycolic acid) (PLGA), developed by COUR Pharmaceuticals. It is uniquely designed to be free of encapsulated drugs or other bioactive agents, exerting its therapeutic effect through the inherent properties of the nanoparticle itself. ONP-302 is designed to target and reprogram myeloid cells, such as macrophages and monocytes. In the context of oncology, it activates the cGAS/STING signaling pathway, which subsequently triggers an IL-15-dependent activation of natural killer (NK) cells and increases the expression of PD-1/PD-L1 within the tumor microenvironment. This immune reprogramming can overcome resistance to checkpoint inhibitors like anti-PD-1 therapy. Additionally, ONP-302 has been investigated for its ability to ameliorate acute inflammatory diseases by modulating myeloid cell activity.
