Overview

Ontamalimab is a fully human immunoglobulin G2 (IgG2) monoclonal antibody that targets mucosal addressin cell adhesion molecule 1 (MAdCAM1). It was developed as a novel treatment for inflammatory bowel diseases by inhibiting the interaction between MAdCAM1 and its ligand α4β7 integrin, thereby reducing lymphocyte trafficking to the gut and modulating intestinal inflammation. Ontamalimab was primarily investigated for moderate to severe Crohn's disease and ulcerative colitis. The drug demonstrated efficacy in inducing and maintaining clinical remission in these conditions but its development program for inflammatory bowel diseases was discontinued by the sponsor for reasons unrelated to safety or efficacy. It has also been studied at an earlier stage for non-alcoholic steatohepatitis[5][6][7].
