Overview

**Onvansertib + cytarabine** is a combination of two pharmaceutical agents used in the treatment of hematologic malignancies, primarily acute myeloid leukemia (AML). Onvansertib is an orally available, highly selective inhibitor of polo-like kinase 1 (PLK1), a serine/threonine kinase essential for proper mitotic progression and cell division. Cytarabine is a nucleoside analog with antimetabolic activity, interfering with DNA synthesis and repair, and is a cornerstone chemotherapeutic agent in leukemia treatment. The combination leverages onvansertib's cell cycle arrest and apoptosis-inducing mechanism with cytarabine's DNA-damaging cytotoxicity to synergistically enhance anti-leukemic activity. Preclinical models and early-phase clinical trials have demonstrated increased survival and antileukemic responses with this combination compared to either agent alone, especially in relapsed or refractory AML and in patients unfit for intensive induction treatment[1][3][5][7]. Onvansertib is developed by Cardiff Oncology (formerly TrovaGene).
