ONYX-015 is a genetically modified adenovirus designed to selectively replicate in and kill cancer cells with p53 mutations. It contains an 827-bp deletion in the E1B region and a point mutation at codon 2022 that generates a stop codon, resulting in loss of function at the E1B locus. The E1B gene product is a 55 kDa protein that normally binds to and inactivates the p53 tumor suppressor protein. This modification allows ONYX-015 to replicate only in cells with impaired p53 function, leaving normal cells unaffected. ONYX-015 has been studied extensively in clinical trials for various solid tumors, particularly head and neck cancers, and has shown enhanced efficacy when combined with chemotherapeutic agents such as cisplatin, 5-fluorouracil, doxorubicin, and paclitaxel.
