OPA894-2 is a preclinical-stage antibody-drug conjugate (ADC) developed by Serilink Biotechnology. It targets B7-H3 (CD276), a transmembrane protein and immune checkpoint member that is frequently overexpressed in various solid tumors, including lung, prostate, and breast cancers, while showing limited expression in normal tissues. The drug is built using Serilink's proprietary OPA (Optimized Payload-Linker) platform, which is designed to enhance the stability of the conjugate in systemic circulation and ensure the targeted delivery of a potent cytotoxic payload, typically a topoisomerase I inhibitor. By optimizing the linker-payload chemistry, OPA894-2 aims to achieve a superior therapeutic index and reduced off-target toxicity compared to earlier-generation ADCs. It is currently being evaluated for the treatment of advanced solid malignancies.
